Bacteriolysis and Variation on the O-side Chain Lengths of Lipopolysaccharides of Salmonella typhi Ty21a with Respective to the Concentrations of Galactose

Abstract
Galactose induced bacteriolysis of Salmonella typhi Ty21a was determined. Notable bacteriolysis of S. typhi Ty21a was observed under 10% galactose, although no bacteriolysis occurred under 0.1% galactose in heart infusion broth. At the same time,
according to the concentration of galactose added to the growth medium, the protein and lipopolysaccharide(LPS) profiles of S. typhi Ty21a in sodium dodecyl sulfate-polyacrylamide gels were examined. With the increase in the concentration of
galactose,
the increase in the length of O-side chain of LPSs of S. typhi Ty21a were prominent. And LPSs extracted from bacteria grown under 1% galactose showed typical ladder-like bands in immunoblot analysis with hyper-immunized rabbit anti-S. typhi Ty2,
which
meant that cells grown under 1% galactose were biosynthesized smooth LPSs with long O-side chains which were similar with LPSs of S. typhi Ty2.
